Summary
To globally evaluate transcriptome treated by decitabine (DAC) in THP-1 and U937 cells expressing p53-R282W, cells were treated with DMSO or DAC. mRNA was isolated and then subject to deep sequencing, using Illumina HiSeq Xten. The sequence reads that passed quality filters were analyzed using htseq. The expressions had been validated using qRT–PCR using SYBR Green assays. Results and conclusions: Global transcriptional change of THP-1 expressing p53-R282W treated with 5 μM DAC was analyzed. Protein interaction network and Enrichment assay was analyzed with the 216 upregulated genes exemplified by ISG15 and IFI27) (Gfold ≥0.4, (RPKM+1 fold change > 1.5), the type I interferon signaling was hit .Similar observations were achieved in U937 cells.
