Summary
This study has successfully established a human FEV reporter system using CRISPR/Cas9 gene-editing technology. We have investigated the precise timing of FEV expression in human SNs. When compared to the previously established TPH2-reporter system, the FEV-reporter system has demonstrated the capacity to yield a greater number of highly-purified SNs, which can be utilized for in-depth exploration of the transcriptomic profiles of SNs during the differentiation process. Moreover, this expanded yield of purified SNs holds promise for facilitating high-throughput drug screening.
