Summary
Aging is a process that all organisms go through. How to delay aging is an urgent problem in the field of life science. The close link between metabolism and aging is well known. In this study, we firstly comprehensively depicted a dynamic metabolic landscape from the oocytes to the cleavage stage embryos, revealing a more reductive characteristic in oocytes. Based on the idea that oocyte-enriched factors promote reprogramming, we demonstrated the multiple roles of an oocyte metabolite 5-hydroxytryptamine (5-HT) in rejuvenation, improving reprogramming efficiency and promoting oocyte maturation. The mechanisms involve a canonical 5-HT function via its receptors and downstream regulation of mitochondrial biogenesis and mitophagy, and a non-canonical function via serotonylation of HSP90AB1. Overall, our study provides a new anti-aging metabolite 5-HT from oocyte-enriched metabolites and helps advance our understanding of rejuvenation strategy through an early embryo development view.
