The response of epidermal nevi to simulated solar radiation using single-nucleus RNA sequencing of frozen tissue

Summary
This study addresses analysis of RNA in single nuclei (snRNAseq) from OCT embedded, frozen human skin/nevus tissue. In this study we created snRNAseq libraries from 6 donors each providing two nevi, one control, and one irradiated with 3-4 times the donor’s erythemal dose (MED) of simulated solar radiation. Both nevi were biopsied 24 hours after irradiation. With no enrichment for rare cell types, our analysis of snRNAseq data reveals the transcriptional diversity of interfollicular and nevomelanocytes, keratinocytes and fibroblast. We also document changes in skin cells, endothelium and cutaneous immune cells elicited by solar simulated radiation.
