Time-resolved transcriptomic responses of kidney distal convoluted tubules induced by furosemide

Summary
The distal convoluted tubule (DCT) of the kidney undergoes tubule remodeling in response to physiologic stresses. This process has been implicated in physiological mechanisms resistant to loop diuretics. However, the molecular mechanisms underlying DCT cell state transitioning in the development of diuretic resistance are unknown. Here, we investigated early cellular responses in the high-resolution transcriptome of the early DCT (DCT1) and an initial activation of cellular regulatory programs after furosemide administration. Time-resolved transcriptomic changes of the DCT1 at four different time points (12h, 24h, 48h, and 96 h) after furosemide administration were identified RNA-Seq analysis in the microdissected DCT1 segments. This study provides comprehensive resource for the early DCT transcriptome and a novel insight into a cellular process of the distal tubule remodeling in the development of loop diuretic resistance.
