Effect of SGC707 on gene expression in SH-SY5Y cells

Summary
The epigenetic regulation of gene expression plays a crucial role in cellular functions and disease progression. In this study, we investigated the effects of SGC707, a selective PRMT3 inhibitor, on the gene expression profile of SH-SY5Y neuroblastoma cells. SH-SY5Y cells were treated with SGC707, and differential gene expression was analyzed using high-throughput RNA sequencing. Combined with CHIP-seq data, we discovered that H4R3me2a modification at the MIR448 promoter promotes the transcriptional activation of miR-448, which in turn suppresses IGF1R expression, ultimately exacerbating Tau phosphorylation. These findings reveal a novel regulatory pathway linking PRMT3 inhibition, miR-448 activation, and Tau pathology, providing potential therapeutic insights for neurodegenerative diseases.
