Generation of functional salivary gland progenitor cells from extended pluripotent stem cells

Summary
The aim of this study was to use pluripotent stem cells (PSCs) as a starting point to progressively differentiate into induced salivary gland progenitor cells (iSGPCs) by mimicking salivary gland development. In this study, we further explored and optimized the passage strategy and organoid formation protocol of iSGPCs based on our previous studies. After 17 days of differentiation, PSC-derived iSGPCs exhibited a marker expression profile similar to that of adult salivary gland progenitors, including KRT5, KRT14, KRT19, and CD49F. Functionally, iSGPCs exhibit self-renewal capacity, are able to form salivary gland-like branching organoids, and have therapeutic potential in models of radiation-induced salivary gland hypofunction.
