Summary
"IFNγ drives antimicrobial responses in TB, mainly via iNOS induction in mouse macrophages, but its role in humans is unclear. We compared IFNγ responses to Mycobacterium tuberculosis across macrophages from multiple species using RNA-seq, bacterial assays, and functional analyses. Unlike mice, human macrophages lacked iNOS induction and failed to restrict Mtb. Only mouse and cattle monocytes showed iNOS activity. scRNA-seq identified airway epithelial cells as potential iNOS⁺ sources in humans.
