Summary
Chromatin profiling with CUT&Tag and CUTAC reveals that while CDK12 inhibition primarily disrupts DNA accessibility at Brg1 and CDK12 co-occupied promoters, BAF inhibition leads to widespread loss of enhancer accessibility. Co-inhibition causes a compounded disruption at shared co-occupied regulatory sites, resulting in loss of chromatin accessibility and transcriptional dysregulation at key loci.
