Summary
Transcriptome sequencing was performed on A375 human melanoma cells with CRISPR/Cas9-mediated knockout of DUS1L and wild-type control cells, with three biological replicates per group. Total RNA was extracted using TRIzol reagent, and RNA quality was assessed by NanoDrop spectrophotometer and Agilent 2100 Bioanalyzer. Indexed RNA-seq libraries were constructed and sequenced on the BGISEQ-500 platform to generate paired-end reads. Clean reads were aligned to the human reference genome (hg38), and gene-level read counts and differential expression profiles were obtained to investigate the impact of DUS1L knockout on global gene expression in A375 cells.
