Summary
Bulk RNA-seq of CRISPR-edited cells identified 742 differentially expressed genes enriched in multi pathways related to cancer, highlighting the widespread regulatory influence of this enhancer. Quantitative PCR validation confirmed consistent expression changes of key genes across multiple mutant cell lines. Collectively, our study demonstrates that GH05J001312 functions as an enhancer with broad impact on oncogenic transcriptional networks, providing a potential therapeutic target in HNSCC.
