Summary
To identify long non-coding RNAs (lncRNAs) associated with Mycobacterium tuberculosis (M.tb) infection, and preliminarily investigate the role of lncRNA ENST00000608721 in macrophages during M.tb infection. Methods: Total RNA was extracted from M.tb-infected and uninfected THP-1 cells, and high-throughput sequencing was performed on the Illumina platform to obtain the transcriptional profile. The differential lncRNAs were subjected to Gene Ontology (GO) analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis, respectively. Furthermore, the differential lncRNAs were verified by real-time quantitative PCR (qPCR). A THP-1 cell model with silenced target lncRNA was constructed using siRNA chemical transfection, and the bacterial load after M.tb infection was determined by colony-forming units (CFU) assay.
