Single-neuron sequencing of PBN-projecting Sp5C neurons in CION or Sham mice

Summary
Parabrachial (PBN)-projecting the caudal part of spinal trigeminal nucleus (Sp5C) neurons are essential for trigeminal neuralgia-induced pain hypersensitivity, but the molecular identities of PBN-projecting Sp5C neurons remain unclear. We have employed single-neuron sequencing by Smart-seq3 to study PBN-projecting Sp5C neurons in chronic constriction injury of the infraorbital nerve(CION)model or Sham mice. This study suggest that the Tac1 gene serve as a specific marker for PBN-projecting Sp5C neurons and demonstrate transcriptional reprogramming induced by CION in PBN-projecting Sp5C Tac1 neurons.
