Summary
We employed fluorescence-activated nuclear sorting (FANS) or Purkinje cells from post-mortem brain tissue of individuals carrying an expanded CAG repeat in ATXN3 gene, a mutation causing spinocerebellar ataxia type 3 (SCA3). Purity of sorted samples was verified by comparing the nuclear transcriptome of Purkinje cell nuclei to the transcriptome of cerebellar granule cell nuclei and glial cell nuclei isolated from unaffected control individuals. The goal was to assess the stability of ATXN3 CAG repeat in pure Purkinje cell samples.
