Summary
We present a spatial Trekker–CUT&Tag multiome protocol that combines direct spatial barcoding of a frozen tissue section with subsequent single-cell multiome analysis. This approach enables integrated single-cell and spatial profiling of histone H3K27ac modification and gene expression, revealing mechanistic and anatomical cell-type organization across the tissue.
