Regulation of gene expression by IL-12P40 heterodimers

Summary
The IL-12 family of cytokines are unique in that they are formed by heterodimerization of two proteins, encoded by distinct genetic loci. We previously reported that the shared subunit of IL-12 and IL-23, namely IL-12P40, can interact biochemically with other partners in vivo. The functional role of these potentially new heterodimeric immunomodulatory cytokines are not known. Here we use hydrodynamic delivery of plasmids encoding four of the top binding partners we identified (CD5L, Resistin-like Gamma, Clusterin and Pentatraxin V) into IL-20-p40 deficient mice, in order to profile the changes in splenic cell gene expression.
