Summary
Synthetic transport vehicles (STVs) are fully engineered, non-viral protein nanoparticles designed to deliver functional RNA cargos with high efficiency while enabling precise control over their molecular composition. Comprehensive RNA profiling demonstrated that STVs package predominantly the intended therapeutic RNA while strongly minimizing the incorporation of endogenous cellular transcripts, highlighting the ability of engineered RNA-protein interactions to direct selective cargo loading. This highly defined RNA composition underpins the robust delivery performance of STVs, enabling efficient transfer of diverse functional RNAs, including mRNAs encoding transcription factors, gene editors, and antiviral effectors, across multiple cell types and in vivo models.
