Effect of SMALS-329 treatment on gene expression in ACHN renal carcinoma cells

Summary
LKB1 is a tumor suppressor kinase whose inactivation is associated with poor prognosis and therapeutic resistance in multiple cancer types. To investigate the transcriptional effects of pharmacological LKB1 activation with SMALS-329, we performed RNA sequencing of ACHN renal carcinoma cells following treatment with 8 mM SMALS-329 for 24 hours. Gene set enrichment analysis identified an enrichment of genes involved in cell adhesion/mobility (11 genes) and cell polarity (6 genes) as the key downregulated group of genes. Additionally, we noted significant regulation, both up and down, of several inflammatory mediators and components of the unfolded protein response (UPR). These findings are consistent with the known effects of LKB1, namely the regulation of metabolism, inflammation, cell adhesion/mobility and cell polarity and the UPR.
