Summary
The communication between tumor-derived exosomes and stroma plays a critical role in facilitating cancer progression. We detected the mRNA expression levels after MRC5 was transfected with shRNAs targeting FOXN3 or incubated with A549 cells-derived exosomes. We reported that in lung cancer microenvironment, tumor cells exhibit a great capacity to convert normal fibroblasts (NFs) to cancer-associated fibroblasts (CAFs). We showed that tumor cells secrete a subset of exosomal microRNAs directly targeting FOXN3, which leads to the activation of fibroblasts. Activated CAFs further promote cancer growth by entering tumor tissues. These results demonstrate that intercellular crosstalk between tumor cells and fibroblasts is mediated by tumor-derived exosomes, providing potential targets for treatment of lung cancer.
