Summary
Cells are exposed to both intrinsic and extrinsic DNA damage stresses in various environmental conditions, necessitating appropriate control mechanisms. Stem cells, characterized by their pluripotency and self-renewal capabilities, must regulate these mechanisms more rigorously due to their high hierarchical status. Previous studies have suggested that stem cells induce differentiation to prevent the propagation of inappropriate DNA damage. However, this approach paradoxically results in the loss of stemness when the DNA damage is repaired. In this study, we aim to demonstrate that stem cells possess mechanisms to protect their stemness in response to DNA damage. To this end, we performed chromatin immunoprecipitation sequencing (ChIP-seq) for Ctbp2, a well-known regulator of stemness escape, and various histone markers.
