Summary
In adult tissues, in response to injury or to the acquisition of a mutational landscape, epithelial stem cells leave their homeostatic state and become activated. To identify the epigenetic factors involved in this process we performed pooled genetic screens integrated with two transcriptional read out systems. These revealed a network of factors including Kmt5c (Suv420h2), Ehmt2 and Cbx3. Through a series of omics and cellular functional assays we discovered their functional role at the cellular and molecular level. In particular we identified a multi-layered control of gene expression, dependent by Cbx3, ranging from transcriptional regulation to mRNA localization.
