Summary
This study investigates the effects of knocking out 17 genes - DMRTA1, TBR1, OVOL1, PITX1, LMO2, ZBTB7C, HEY1, MBNL2, DLX4, DLX5, DLX6, HOPX, MAFF, TWIST2, MBNL3, SIX3 and NR2F1, by inserting premature termination codons (PTCs) in primitive syncytium and cortical brain organoid model systems derived from the KOLF2.2J iPSC line. Bulk RNA-seq analysis was performed to examine the transcriptional consequences of these gene knockouts, providing insights into their roles in brain development and cellular differentiation in organoid models
