Impact of METTL1 on global gene expression in AML

Summary
Methyltransferase 1 (METTL1) is an enzyme responsible for the m7G modification of tRNAs. Our studies indicated that METTL1 is highly expressed in AML samples and its elevated level predicts poor prognosis of AML patients. Knockout (KO) of METTL1 in AML cells significantly suppressed their growth, promoted myeloid differentiation in vitro, and substantially delayed leukemogenesis in vivo. Additional rescue assays demonstrated the functions of METTL1 in AML primarily depended on its enzymatic activity. To delineate the molecular mechanism through which METTL1 exerts its biological functions in AML, we conducted transcriptome-wide RNA sequencing with Mono-mac-6 AML cells following METTL1 KO.
