Effects of WDFY3 overexpression on the transcriptomic profile of murine bone-marrow derived macrophages.

Summary
We determined the transcriptomic profile of bone-marrow derived macrophages (BMDMs) from mice overexpressing human WDFY3 and their wild-type littermate control mice. Specifically, these mice were engineered by knocking a construct consisting of the cDNA sequence encoding a 3x FLAG tag fused to human WDFY3 cDNA, preceded by a STOP cassette flanked by loxP sites, into the Rosa26 locus (Rosa26 loxP-STOP-loxP-3xFLAG-hWDFY3/+), with expression driven by the endogenous Rosa26 promoter. To achieve constitutive overexpression, male mice carrying the floxed allele were crossed with female carriers of the HprtCre strain (Stock No. 004302, The Jackson Laboratory), a Cre-deleter strain with 100% Cre-mediated recombination in oocytes, to generate heritable deletion of the loxP-STOP-loxP cassette (Rosa26 3xFLAG-hWDFY3/+). These Rosa26 3xFLAG-hWDFY3/+ mice were interbred to produce homozygotes (Rosa26 3xFLAG-hWDFY3/3xFLAG-hWDFY3) for knock-in (KI) of 3xFLAG-hWDFY3 (hWDFY3KI/KI) and wild-type littermate controls (Ctrl).
