NOP16 regulates the B cell class switch recombination into IgA

Summary
Class switch recombination (CSR) is a DNA recombination process in B cells that generates diverse isotypes, playing a crucial role in humoral immune responses. However, the regulatory mechanisms underlying CSR remain incompletely understood. In this study, we investigated the role of the nucleolar protein NOP16 in B cell CSR. Using the mouse B cell lymphoma cell line CH12F3-2A, we induced class switching from IgM to IgA via stimulation with CD40L, IL-4, and TGF-β. Compared to wild-type cells, CH12F3-2A cells with NOP16 knockout via CRISPR-Cas9 exhibited a significant enhancement in IgA class switching. To elucidate the transcriptional differences between wild-type and NOP16-deficient cells during IgA CSR, we performed transcriptome analysis by bulk RNA sequencing (RNA-seq).
