Lineage Tracing Reveals a Common Ancestor of Supraclavicular Brown and Inguinal Beige Adipocytes

Summary
Like human scBAT, supraclavicular brown adipose tissue (scBAT) consists of loosely connected thin strips of brown adipocyte islands and is located in the mouse neck. We traced the origin of scBAT to Mef2c-anterior heart field (AHF)-marked (Mef2c-AHF⁺) cells. To investigate the differentiation potential of these cells, we isolated and immortalized Mef2c-AHF⁺ cells (named Mef2c-AHF-P5 cells) from mouse scBAT. Using this cell line, we performed RNA sequencing to profile gene expression induced by either standard adipogenic induction medium (IM) or a combination of Bmp7 and Rosiglitazone. Our results indicate that IM and the Bmp7-Rosiglitazone (BR) combination induce distinct gene expression profiles during the early differentiation of Mef2c-AHF⁺ cells.
