High Expression of TMEM161A in Colorectal Cancer is Associated with Tumor Proliferation and Poor Prognosis

Summary
TMEM161A is a membrane-associated protein implicated in oxidative stress responses, but its role in colorectal cancer (CRC) has not been elucidated. We analyzed 133 CRC specimens and found that high TMEM161A expression correlated with poor prognosis. siRNA-mediated TMEM161A knockdown in CRC cell lines (HT29, LIM1215) suppressed proliferation, colony formation, and invasion, with delayed G2/M transition. RNA-seq and GSEA revealed downregulation of E2F targets, G2/M checkpoint, MYC, and mTORC1 signaling, and upregulation of p53 and TNFα–NFκB pathways. TMEM161A thus promotes CRC progression and may serve as a prognostic biomarker and therapeutic target.
