Summary
Virtual memory CD8+ T (TVM) cells are a distinct subset of antigen-inexperienced cells with rapid effector potential, but their regulatory checkpoints remain elusive. Here, using single-cell RNA sequencing and single-cell ATAC sequencing, we found that loss of RPA1 promotes chromatin remodeling in CD8+ T cells, leading to enhanced accessibility at EOMES-associated loci and activation of transcriptional programs that drive TVM cell polarization.
