Summary
We profiled transcriptomic changes in SW48 cells after 5-day 100μM levofloxacin treatment. Levofloxacin induces DNA damage and moderate p53 activation biased toward DNA repair over pro-apoptotic programs, while remodeling mitochondrial metabolism via enhanced OXPHOS and ROS accumulation. Though levofloxacin alone does not trigger robust apoptosis, it primes cells for apoptotic sensitivity via metabolic stress and redox imbalance, defining a novel anti-tumor mechanism of this clinical antimicrobial.
