Summary
Purpose: Previous studies demonstrated that BCAA metabolic reprogramming induces idiopathic pulmonary fibrosis (IPF). This study investigates BCAA-mediated gene expression regulation in primary mouse lung fibroblasts (MLFs). Methods: MLFs were cultured in two media conditions: complete medium (Complete) or BCAA-free medium (Free) for 24 hours prior to TGF-β1 (20 ng/mL) stimulation for 48 hours. H3K4me3, H3K27ac, H3K27me3, H3K36me3, and H3K9me3 CUT&Tag analyses were conducted on treated MLFs. Conclusions: BCAA plays a crucial role in modulating H3K36me3 of key fibrotic genes.
