Summary
To investigate the effect of hepcidin-1 deficiency on gene expression of mouse hepatic Kupffer cells, we isolated hepatic non-parenchymal cells from hepcidin-1 knockout mice (KO) and wild type (WT) mice with C57BL/6 genetic background, and then obtained fluorescence-activated cell sorting (FACS) sorted-Kupffer cells (gated on live CD45+ F4/80+ CD11bint hepatic nonparenchymal cells). We then performed switching mechanism at the 5′ end of the RNA transcript (SMART-seq) analysis of isolated Kufpffer cells.
