RNA Sequencing of Lung Tissue in FTO-Overexpressing Mice Modeling Chronic Obstructive Pulmonary Disease

Summary
Fat mass and obesity-associated protein (FTO) is linked to various diseases; however, its role in COPD remains unclear. We previously found that the expression level of FTO was significantly decreased in COPD models. To explore the downstream target genes and pathways of FTO, we conducted RNA sequencing on lung tissues from wild-type and FTO-overexpressing mice exposed to cigarette smoke. Here, we report that FTO plays a crucial role in regulating inflammation in COPD. We demonstrated that FTO expression can modulate several pathways, including the PI3K-Akt and MAPK signaling pathways, which are associated with inflammation. Transcriptomic analysis further revealed FTO-dependent alterations in genes associated with inflammation. Overall, our study enhances the understanding of the molecular mechanisms by which FTO contributes to the regulation of inflammation in COPD.
