Summary
Using 10x Genomics single nuclear RNA sequencing (snRNA-seq) to characterize differences between mice with and with Brca1 specifically in the liver, after carcinogen exposure. Hepatocellular carcinoma (HCC) is among the most common and deadly cancers worldwide and HCC-associated cases and deaths are projected to increase. Recently, tumor suppressor gene Breast cancer susceptibility gene 1 (BRCA1) was implicated in liver tumor development, yet its mechanisitc role remains unclear. We found that hepatic Brca1 deficiency in mice paradoxically protected against carcinogen induced liver tumor development. Using snRNAseq, we sought to characterize differences in non-tumor liver tissue to determine the underlying mechanism driving liver tumor protection.
