Transcriptomic determinants of humoral immunity for heterogeneously antigenic PL45 cells

GSE334534 Homo sapiens Expression profiling by high throughput sequencing 12 samples 2026/06/12 GPL34284
Summary
Ascitic fluid from patients with pancreatic ductal adenocarcinoma (PDAC) contains IgG antibodies that bind to allogeneic PDAC cell lines, including the commercially available PDAC cell line PL45. PL45 cells exhibited heterogeneous binding by IgG from patients with PDAC (A1 and A14). We hypothesized that transcriptomic heterogeneity within the cell line contributes to the observed antigenic heterogeneity. To investigate potential transcriptomic determinants of humoral antigenicity, PL45 cells were stained with ascitic IgG from patients A1 or A14 and flow-cytometrically sorted using a fluorescently labeled anti-human IgG detection antibody. RNA was isolated from the IgG-positive and IgG-negative PL45 subpopulations and subjected to bulk RNA sequencing followed by DESeq2 differential expression analysis.
