Summary
To determine the key transcription factors and molecular pathways involved in Limbic Encephalitis (LE), we analyzed immune cell-driven transcriptional changes in a newly developed mouse model of CD8+ T cell-mediated LE. This murine model (OVA-CD8+ LE) recapitulates the essential phenotypic features observed in human cytotoxic T lymphocyte (CTL)-induced LE, including neuroinflammation and seizure susceptibility. In a time series approach 2, 5, 8 and 28 days post-injection hippocampal tissue was harvested for RNA sequencing analysis. These timepoints were selected to capture stage-specific transcriptional changes driven by CD8⁺ T cell activity, thereby providing insights into the molecular events underlying hippocampal dysfunction in LE.
