Summary
This study investigates how pharmacologic activationn of the PERK branch of the unfolded protein response (UPR) enhances the immunoregulatory function of FoxP3⁺ regulatory T cells (Tregs) in the context of intestinal inflammation. Using ER stress-inducing agents and PERK activators, combined with flow cytometry, bulk RNA sequencing, in vitro Treg conversion and suppression assays, ELISA, qRT-PCR, and immunoblotting, we demonstrate that PERK activation reduces gut inflammation and promotes Treg-mediated immune regulation. These findings identify the PERK pathway as a novel immunologic target for enhancing Treg function and mitigating chronic intestinal inflammation, offering translational relevance for therapeutic strategies in inflammatory bowel disease (IBD).
