Summary
Current strategies for therapeutic use of mRNAs and for testing the function of specific mRNA isoforms assumes that transfected mRNAs are not further processed by cellular splicing machinery. Here we show that lipofected mRNAs can be spliced in human cells, utilizing canonical splice donor and acceptor sites and this is suppressed by spliceosome inhibition. This post-transcriptional splicing has important strategic implications for the use of transfected mRNAs in experiments and therapies.
