Summary
Further study on the mechanism of glucocorticoid (GC) -induced apoptosis of osteoblasts (OB) is of great practical significance for the prevention and treatment of steroid-induced osteoporosis and osteonecrosis. However, the regulatory mechanism of alternative splicing in GC-induced OB apoptosis remains unclear. The aim of this study is to explore the role and mechanism of the splicing regulator serine/arginine-rich splicing factor 1 (SRSF1) in GC-induced OB apoptosis. OBs were treated with 0.4mg/ml hydrocortisone, the active form of GC, for 12 hours, and the mRNA expression profile was obtained by RNA-seq. The key splicing factor SRSF1 mediating GC-induced OB apoptosis was successfully screened, and its role and regulatory mechanism were further determined. Importantly, overexpression of SRSF1 was resistant to GC-induced OB apoptosis.
