Summary
In this present study we demonstrated portal hypertension after PHx promoted liver regeneration by activating mechanoreceptor Piezo1 in endothelial cells. Next, in vivo and in vitro experiment proved that Piezo1 activation in LESCs enhanced liver regeneration via Wnt2/β-catenin mediated LESCs-hepatocyte crosstalk: Shear stress-induced Piezo1 activation in LESCs promoted secretion of Wnt2 protein to the liver microenvironment via AFT2/ID1 signaling pathway, which further promoted the nuclear translocation of β-catenin in hepatocytes and consequently enhanced the proliferation of hepatocytes (Fig. 6M). In addition, we confirmed the translational value of Piezo1 agonist Yoda1 in promoting liver regeneration after PHx.
